Morphological and Molecular Characterization of Gnetum africanum (Welw.) Germplasm Using DNA Barcoding Method

چکیده

Gnetum africanum is a twisted woody climbing wild plant i.e.liana found growing naturally in dense equatorial rainforest but currently domesticated tropical areas of the world. There has been taxonomic confusion classifying this liana due to its observable diverse morphological characters. Indeed, characterization alone not offered needed solution. However, there dearth information on molecular africanum. Morphological and descriptors, could help offers phenotypic genotypic much better. This study aimed at characterizing land races identify species that be used for development high yielding hybrids. Three different or accessions was collected from locations Nigeria. High quality genomic DNA isolated samples; it checked purity quantified as appropriate. The samples were subjected PCR amplification barcoding studies (chloroplast large subunit ribulose -1, 5-bisphophate carboxylase rbcL gene). amplicons gel eluted, sequenced homology by using Basic local alignment search tool -BLAST. Identification obtained top similar outcomes samples. For Ekim Fig. 1b sample, forward blast showed 95% similarity with Basella alba voucherID: NC041293.1 reverse had 88% Talinum fruticosum cultivar ID: MK598685.1. Etinan 1a 86% Rhabdothamnussolandri voucher JQ933464.1 reverse, no founded.
